Subject: is the matter with which the work will be dealing.

Theme: is a commentary on the subject matter.

example:

Subject: Racism.

Theme: Racism is neither good nor evil but simply a commentary on statistical facts about human genetics. It is the utilization of these facts and statistics by human beings that determines its 'evil', 'goodness' or 'mundanity'.
