There are chief gods and goddesses that play a role on Mythology Island.
- Zeus - "king of the gods" is seen as upset with sharing power with his "brothers" Hades and Poseidon, to the point of writing graffiti on their temples and scheming to seize total power
- Athena - daughter of Athena, but opposes his plans. The olive branches (peace) are links to her helpful advice.
- Hercules - the demi-god is offering Autographs in his food court. It is not until Zeus threatens to control all of Poptropican Greece that he decides to help
- Hades - ruler of the Underworld. His monster guard dog Cerberus holds one of the Sacred Items (a whisker)
- Poseidon - ruler of the sea. His realm contains the giant pearl and the Hydra cave (a scale from the Hydra is one of the Five Items).
- Aphrodite - goddess of beauty. She is in the realm of Poseidon and offers you a useful tool, a Mirror that can transport you throughout the four realms.
Other mythological individuals and creatures : a satyr, the Minotaur, the Sphinx (Greek legend), the Hydra, and Cerberus, guardian of the underworld.
