When installing asphalt shingles should the cellophane paper over the tar strip be removed?

You can, but you do not have to. That is there to keep them from sticking together when in the bundle. Removing it will let the shingle stick to the felt and may produce a roof that is more of a solid sheet, but it is not going to make a big difference either way.
