First and most importantly, The Diary of Anne Frank is not a novel. Novels are a work of fiction; Anne Frank's journal was a factual account of her everyday life.
As far as major themes go, there are several:
- The idea of being discriminated against for no other reason than one's ethnic and religious background,
- The psychological toll of having to remain quiet and hidden, fearing the terrible consequences of discovery (which history unfortunately proved out),
- And the attempt to have as normal a day-to-day existence as possible, given the above factors.
