This is very funny that somebody is asking this. Obviously someone has been listen to Waka Flocka Flame too much and is trying to understand his lyrics. Run up in yo spot is basicallly saying go into you trap or your spot like where you chill at and take all your valuables. Drugs dealers and gang afilatated people are usually involve with this term.
