The Port Authority Bus Terminal (42nd Street at 8th Avenue) is the terminus for Greyhound buses. You can walk 1 block east on 42nd Street to 7th Avenue, and enter the subway at the 42nd Street-Times Square station, or you can enter the subway system at 42nd Street-Port Authority, and then walk through the tunnel that connects the 42nd-Port Authority station to the 42nd-Times Square station.
Take the Uptown 7 train (the purple line), 1 stop, from 42nd Street-Times Square to 42nd Street-Bryant Park. Transfer at Bryant Park to the Uptown F train (of the B-D-F-M, the orange line). Take the Uptown F to the end of the line, the Jamaica-179th Street station.
If the 7 train isn't running, you can just walk to the 42nd Street-Bryant Park station. It's only 2 blocks (not counting Broadway, which cuts between avenue blocks): 8th Avenue ---> 7th Avenue ---> 6th Avenue.
If you can't walk because you have too much luggage, then you can take the Downtown A-C-E (the blue line) from 42nd Street-Port Authority to West 4th Street, and transfer at West 4th Street to the Uptown F.
