You can determine what a star is made of by close examination of the light that it emits. Every different substance emits a different combination of light frequencies when it is heated to the point at which it glows. You can pass light through a prism and break it up into its component frequencies (or colors) and analyse them in great detail, with the right kind of equipment and training.
