Why the Battle of Shiloh was the bloodiest battle so far in the Civil War?

Mainly because it was the first big battle of the war, and the armies had had time to build-up. (Bull Run looked big at the time, but it would be classified as a small battle in context.)

Also, Grant was caught off-guard, so the casualties were bigger than they ought to have been.
