The answer is Go because it has an implied you in front of it. Only commands can have an implied you and no is not a command. So basically the whole sentence is (You) Go.

There are several one-Word sentences in English that immediately come to mind. So if you mean shortest as in the nomber of sords, there are quite a few. For example:

But if you mean the shortest as in the least amount of letters used, the shortest I can think of is No.

(As in:

"Do you want rat poison, cyanide, or glass shards in your caviar this evening, madam?" said the crazy cook.
