When you are perfectly sure you are gay, and you know enough about it to be able to answer some of their worries.

Maybe tell one of them first, not both together? Choose a quiet time when you are with them. Don't announce it at the Christmas or Thanksgiving dinner table!

There are ways you can prepare them, if you think it's a good idea. Get some literature from PFLAG (Parents and Friends of Lesbians and Gays. Check online), and leave it lying around.

Remember if they haven't suspected it, then they might be in shock or denial, and they might say things without thinking. ("No you're not!" "Get out of my house!" "That's impossible!" "Not my son!" "We'll get a doctor to fix you..." etc). If they say something in haste, then don't argue with them. Go away and let them think about it for a while.
