To inoculate Triple Sugar Iron (TSI) agar, use a sterile inoculating loop to obtain a small amount of the bacterial culture. Streak the loop across the surface of the slant in a zigzag pattern, then stab the loop into the butt of the tube to ensure anaerobic conditions. After inoculation, incubate the tube at 35-37°C for 18-24 hours to allow for bacterial growth and color change. Finally, observe the results for carbohydrate fermentation and gas production.
