Most states require some on the job experience before you can even apply for the license. I would suggest some time hitting the books, math (algebra is a must) basic chemestry, as well as knowing the state laws and codes. Then one must take a test to prove he has the knowledge to operate a water (or wastewater) plant. If you pass, the state will issue a certification as a water plant operator. A little college helps, classes like microbiology, enviromental Biology, math, and chemestry.
