Solid sequencing, or sequencing by oligonucleotide ligation and detection, involves synthesizing short DNA fragments and determining their sequences through a series of ligation and detection steps. In this method, a template DNA is hybridized to a set of fluorescently labeled oligonucleotides, which are sequentially ligated to the template based on complementary base pairing. After each ligation step, the incorporated fluorescent signal is detected, allowing the sequence of the DNA template to be determined. This process is repeated multiple times to generate longer sequence reads.
